What's Tongkat Ali
Eurycoma longifolia commonly known as tongkat ali, Malaysian Ginseng, or Longjack, is a tall, slender evergreen flowering shrub-tree in the family Simaroubaceae.This plant is commonly found on sandy soil in Southeast Asia including Myanmar, Thailand, Laos, Cambodia, and Malaysia.In vitro studies have been carried out on the plant's antimalarial, antiulcer, antipyretic, and aphrodisiac activity as well as cytotoxic activity against cancer cells. The tea made from this plant is popularly taken as a traditional remedy to improve the male libido, sexual prowess and fertility. Presently, many tea, coffee and carbonated beverages, pre-mixed with the root extract are available commercially for the improvement of general health and labido. As a result, this plant is in such high demand that it is now considered a “protected” species.
Main Active Constituents
The extract of the roots has been found to contain quassinoids (e.g. eurycolactones A-F, eurycomalides A-B, eurycomalactone, 6α-hydroxyeurycomalactone, 6-hydroxy-5,6-dehydroeurycomalactone, 5,6-dehydroeurycomalactone, eurycomanone, 13β,21-dihydroxyeurycomanol, 14,15β-dihydroxyklaineanone, 5α,14β,15β-trihydroxyklaineanone, laurycolactones A-B, longilactone, 6-dehydroxylongilactone, 2,3-dehydro-4α-hydroxylongilactone and pasakbumin B-C), canthin-6-one alkaloids (e.g. canthin-6-one, 1-hydroxycanthin-6-one, 9-hydroxycanthin-6-one, 5-hydroxymethylcanthin-6-one, 5-methoxycanthin-6-one, 9-methoxycanthin-6-one, 10-methoxycanthin-6-one, 1-hydroxy-9-methoxycanthin-6-one, 4-hydroxy-5-methoxycanthin-6-one, 8-hydroxy-9-methoxycanthin-6-one, 5-hydroxymethyl-9-methoxycanthin-6-one, 4,5-dimethoxycanthin-6-one, 9,10-dimethoxycanthin-6-one, canthin-6-one 9-O-β-glucopyranoside, canthin-6-one-3N-oxide, 9-hydroxycanthin-6-one-3N-oxide and 9-methoxycanthin-6-one-3N-oxide), β-carboline alkaloids (e.g. β-carboline-1-propionic acid, 7-methoxy-β-carboline-1-propionic acid, methyl β-carboline-1-carboxylate, n-pentyl β-carboline-1-propionate and picrasidine L), triterpenes (e.g. eurylene, mixture of β-sitosterol and stigmasterol, and β-sitosteryl glucoside) and others (e.g. scopoletin, fraxidin, scopolin, ρ-hydroxybenzaldehyde, syringic aldehyde, 2,4’-dihydroxy-3’-methoxyacetophenone, 2,3-dihydroxy-1-(4’-hydroxy-3’-methoxyphenyl)-propan-1-one, 3-hydroxy-1-(4’-hydroxy-3’-methoxyphenyl)propan-1-one, threo-1,2-bis-(4-hydroxy-3-methoxyphenyl)propane-1,3-diol, vanillic acid, protocatechuic acid, nicotinic acid, syringic acid, sodium syringate, sodium ρ-hydroxybenzoate, lariciresinol, erythro-1-C-syringylglycerol, threo-1-C-syringylglycerol, erythro-guaiacylglycerol, threo-guaiacylglycerol, iandonone, adenosine, guanosine, thymidine, alanine, proline, arginine, serine, glucose and fructose).
There are numerous reviews and phytochemical studies on Eurycoma longifolia. The plant's pharmacological activity is attributed to various quassinoids icluding eurycomanone and eurycomanol, squalene derivatives, biphenylneolignans, tirucallane-type triterpenes, canthine-6-1, and beta-carboline alkaloids.
Eurycomanone is one of the oxygenated quassinoids isolated from Eurycoma longifolia .It is found to be involved in phytochemical activities of eurycoma longifolia including testosterone steroidogenesis and spermatogenesis increase.
Problem in Tongkat Ali Extract Industry
Tongkat Ali Extract products stating various ratios are common on the market. However, according to the New York Times and Washington Post, in 2015, the New York attorney general examined major brand supplements and found them to be mostly fillers. In Malaysia, more evidences showed that more than 41% of the commercially available Tongkat Ali product samples contained no eurycomanone and 24% contained measly eurycomanone.
The fundamental cause of this issue is because currently there is no unified standardized qualification criteria in the industry so that it is impossible to verify the genuineness of the product. As a result, there are many Tongkat Ali products are found be fake which contained no eurycomanone at all.
OHI standard Tongkat Ali Extract product
To address this condition OHI studied and developed standardized Tongkat Ali extract. A standardized extract is an extract that contains a concentrated and set percentage of active ingredients that has been HPLC tested. In the case of Tongkat Ali, eurycomanone is the most commonly studied constituent. Therefore, OHI provides HPLC tested Tongkat Ali extract with standardized Eurycomanone to ensure the authenticity and potency of the product.
Some brands will claim ridiculous amounts of eurycomanone such as 25%, but their legitimacy are highly questionable as eurycomanone 25% extracts are normally only available from R&D laboratories which cost more than 10,000 dollars per kilo and will lose many other beneficial constituents in Tongkat Ali. OHI supplies standardized Tongkat Ali extract that comes up to the legal required standard in Malaysia (eurycomanone 0.8-1.5 w/v (%)) in order to guarantee the genuineness as well as the overall quality of the product.
Eurycoma longifolia Jack(ELK) is popularly taken as a traditional remedy to improve the male libido .The effects of ELK extract were studied on the initiation of sexual performance in rats.A study investigated ELK efficiency on improving sexual performance of inexperienced castrated male rats.The ELK oral administration showed a dose-dependent increase in sexual performance of the treated animals which is supported by other studies.In 2003 Phytomedicine published a further exploration of EJK aphrodisiac activity .The study determined EJK enhanced the sexual qualities of the middle aged male rats by decreasing their hesitation time as compared to controls with various fractions of EJK produced.The study shows that E. longifolia Jack enhanced the sexual qualities of the middle aged male rats, further supports the folkuse of EJK as an aphrodisiac.
Researches showed isolation from Eurycoma longifolia Jack(ELK) ,Eurycomalactone ,prolonged the survival time of mice infected with Plasmodium berghei at a concentration of 2.5 mg/kg. However, this concentration caused early death in the test animals. Additional experiments using a 34% alcoholic extract, water extract, n-hexane fraction, chloroform fraction, and 95% ethanolic fraction also proved toxic to the test animals.In vitro antimalarial activity was found against P. berghei at an inhibitory concentration of 4.5 × 10 −7 g/mL.It is suggested that EJK extract Eurycomalactone may developed in a novel measure in antimalarial treatment.
Antiulcer and antitumor activity
Eearly investigation of antiulcer quassinoids from Eurycoma longifolia was published by European Journal of Medicinal Chemistry.The study revealed both pasakbumin-A (= eurycomanone) and pasakbumin-B which are generally recognised as antitumor compounds were revealed to have antiulcer activity.Later studies concluded Eurycoma longifolia possesses potent anti-tumor promoting activities due to the occurrence of a variety of quassinoids.Further investigation reported on Anticancer Res evaluated the Eurycoma longifolia root extract activities against the growth of a human breast cancer cell line, MCF-7.
The study result indicated that Eurycoma longifolia extracts and fractions exert a direct antiproliferative activity on MCF-7. The resultant from Eurycoma longifolia extracts purification(ELEP), exhibited a higher cytotoxic activity towards MCF-7 by significantly increased apoptosis in MCF-7 cells.Moreover ,ELEP did not affect the expression of the pro-apoptotic protein, Bax. These results, therefore, suggest that ELEP has antiproliferative effects on MCF-7 cells by inducing apoptosis through the modulation of Bcl-2 protein levels.
The anxiolytic effect of Eurycoma longifolia Jack in mice was examined .According the early investigation on Japanese Journal of Pharmacology ,Eurycoma longifolia jack extract (EJK) possess consistent anxiolytic effect with diazepam therefore may applied as an alternative treatment for anxiety therapy.
Blood glucose decreased in streptozotocin-induced hyperglycemic adult rats after treatment of 150 mg/kg body weight using aqueous extracts of E. longifolia . Blood glucose levels decreased 38% ( p < 0.05) and 47% ( p < 0.001) for 2 different extracts.
Improve immune responses
Latest research showed Eurycoma longifolia possess potential to enhance immune functions .Human study on International Journal of Sports Science indicated natural killer (NK) cells count was significantly higher (P = 0.02) at the end of the EL trial compared to the P trial (4.8 and 4.0 x 103.dL-1 respectively). Study result suggested that Eurycoma longifolia (EL) supplementation may improve immune responses following endurance running in the heat.
